Secretion of Cytokine Stimulating Intercellular Adhesion Molecule-1 Expression from THP-1 Cells Infected with Human Cytomegalovirus

Abstract
Human cytomegalovirus (HCMV) stimulates the expression of intercellular adhesion molecule (ICAM-1) on
the surface of monocytic THP-1 cells. Stimulation of ICAM-1 did not require HCMV gene expression since
UV-inactivated HCMV (UV-HCMV) was able to induce ICAM-1 expression. ICAM-1 expression was also
stimulated in uninfected THP-1 cells which were fed with culture supernatant of HCMV-infected THP-1 cells.
Co-culture experiment using trans-well insert supported that HCMV-infected THP-1 cells secreted some cytokine(
s) stimulating ICAM-1 expression. The stimulation of ICAM-1 by HCMV-infected cell culture supernatant
appears to involve NF-¥êB pathway. Culture supernatant from THP-1 cells infected with UV-HCMV, whose
gene expression was abrogated, failed to stimulate ICAM-1 expression on naive THP-1 cells. Thus, HCMV
gene expression seems to be required in secretion of cytokine(s) stimulating ICAM-1 expression.
